Minovia Therapeutics Reports First Clinical Data Demonstrating Disease-Modifying Efficacy and Safety of Mitochondrial Augmentation Therapy in Pediatric Patients with Primary Mitochondrial Diseases

First in human clinical study provides proof-of-concept for mitochondrial augmentation therapy platform, which enables use of healthy mitochondria to improve mitochondrial function and mitigate effects of large-scale mitochondrial DNA (mtDNA) deletion syndromes

No treatment-related adverse effects reported in study

WOBURN, Mass. and HAIFA, Israel, Dec. 21, 2022 (GLOBE NEWSWIRE) — Minovia Therapeutics, a clinical-stage global biotechnology company, today announced that the findings of the first clinical use of mitochondrial augmentation therapy (MAT) platform to treat pediatric patients with primary mitochondrial diseases have been published in Science Translational Medicine. The study was conducted in collaboration with global leaders in the field of hematology and primary mitochondrial disease at Sheba Medical Center (Tel Hashomer, Israel).

Single, large deletions in mitochondrial DNA (mtDNA) can lead to a variety of devastating diseases, including Pearson Syndrome and Kearns-Sayre Syndrome (KSS). These mtDNA deletion syndromes are sporadic, uncurable and ultimately fatal. Pearson syndrome is a bone marrow failure disease that usually begins in infancy. In addition to presenting with sideroblastic anemia, patients are characterized by exocrine pancreas dysfunction. About half of patients die in infancy or childhood, while many who survive go on to develop KSS, a progressive multisystem disorder.

“These findings demonstrate that mitochondrial augmentation therapy is feasible in children with mitochondrial DNA deletion syndromes and that autologous CD34+ cells augmented with mitochondria derived from maternal blood may potentially deliver some functional improvement for patients living with these debilitating diseases for which there are no available therapies,” said Elad Jacoby, M.D., Ph.D., Hemato-oncology Pediatric department, Sheba Medical Center.

MAT is designed to rescue mitochondrial function and metabolic activity in diseased cells through enrichment with healthy mitochondria. This builds upon Minovia’s previous demonstration that MAT provides long-term functional benefit in an immunocompromised mouse model, and that mitochondria can transfer between hematopoietic stem cells (HSCs) in vivo (Jacoby et al 2021). Preclinical studies showed that hematopoietic stem and progenitor cells (HSPCs) can transfer normal mitochondria to hematopoietic and non-hematopoietic cells and improve disease features (Rocca et al., 2017).

Minovia is developing MAT to treat Primary Mitochondrial Diseases such as Pearson syndrome, as well as other mitochondrially-related sideroblastic anemias. The Company’s lead investigational candidate, MNV-101 (MNV-BM-BLD), has been granted the Fast-Track, Orphan Drug and Rare Pediatric Disease designations by the U.S. Food and Drug Administration.

The latest study reported results from six patients ranging from 2 to 7 years of age: four with Pearson syndrome and two with KSS spectrum. All six patients suffered from significant failure to thrive and advanced multi-organ disease, which was either immediately or potentially life-threatening, requiring multiple ongoing supportive care interventions.

Key Findings

  • No treatment-related adverse events were reported. Adverse events related to the leukapheresis procedure were managed with standard medical care. Adverse events that occurred following treatment were expected due to the patient’s underlying disease and resolved spontaneously or with appropriate treatment.
  • Higher levels of normal mitochondrial DNA were observed in the blood of four out of six patients, indicating a reduction of mitochondria with large DNA deletions.
  • Clinical improvement in aerobic function was observed in some patients, and five of six patients experienced weight gain.
  • Quality of life measurements showed improvement in most patients. Changes in the general well-being of children and physical activity were reported, along with increased time spent awake and in play.
  • A series of aerobic and endurance tests were performed on two patients. Both patients demonstrated improvement in leg muscle strength and endurance at 6 and 12 months post treatment.
  • One patient, who at baseline was unable to walk any distance, was able to walk 10 meters at 12 months and had improvement on the 30-second sit-to-stand test.
  • Another patient demonstrated improvement in muscle function in some tests and a sustained improvement in others, including the six-minute walk test and sit-to-stand capacity at 6 and 12 months.

Onecity Expo 2022

 

The Onecity Expo 2022 is also taking place until the 1st of January 2023. The 3rd expo organised by Onecity Event and Marketing management is in conjunction with the Brunei December Festival 2022.

Khat Competition Prize Presentation

Various ‘khat’ or Islamic calligraphy programmes and activities have been lined up by the Islamic Calligraphy and Art Study Centre, Yayasan Sultan Haji Hassanal Bolkiah throughout the year, including a Khat Competition aimed to instil interest towards khat and polish the people’s talents and skills in khat. This year, the centre held the ASEAN Level Khat Competition for the very first time to commemorate Yayasan’s 30th anniversary of its establishment. Yesterday afternoon, the ASEAN Level Khat Competition 2022 Prize Presentation Ceremony was held at Dewan An-Naura, Yayasan Sultan Haji Hassanal Bolkiah Complex.

The 6th Brunei Film Blitz Festival launched the 96-Hour filmmaking competition yesterday morning. Held in conjunction with the Brunei December Festival 2022, 27 participants are taking part this year, doubled from last year’s participation.

 

Winners of the Brunei Film Blitz competitive categories will be awarded at the Screening and Gala at the end of this month. Dayang Siti Kamaluddin, the Festival Director noted the vast improvement of film entries, and the tremendous improvement of quality of films produced. The festival acts as a platform for many to network and launch their career in film.

BN on the Move Prize Presentation

 

BN on the Move, an ongoing campaign organised by the Ministry of Health for the health promotion plan has launched the 20 Billion Steps Together Challenge starting October to the end of this year. To date, over 70,000 users are joining the challenge, with over 17 billion steps recorded so far. Yesterday afternoon, a prize presentation ceremony was held for the winners of the 20 Billion Steps Together Challenge – BN on the Move campaign for November at the Atrium of the Health Promotion Centre.

BTFA Celebration 2022

The Democratic Republic of Timor Leste in collaboration with the Brunei Timor-Leste Friendship Association, organised the 1st Brunei Darussalam and Timor-Leste Friendship Association, BTFA Celebration 2022. The event took place yesterday morning at the Embassy of Timor-Leste in Jalan Kebangsaan.

 

In attendance was Pengiran Mohammad Amirizal bin Pengiran Haji Mahmud, Permanent Secretary for Sports at the Ministry of Culture, Youth and Sports. In his speech, His Excellency Abel Guterres, Ambassador Extraordinary and Plenipotentiary of the Democratic Republic of Timor Leste stressed that this year marks the 20th anniversary of the establishment of diplomatic relations between both countries in the areas of cultural and sport activities.

Philippines Orders Strengthened Military Presence After ‘Chinese Activities’ Near Islands

 

The Philippines’ defense ministry on Thursday ordered the military to strengthen its presence in the South China Sea after monitoring “Chinese activities” in disputed waters close to a strategic Philippine-held island.

The ministry did not specify what activities those were and its statement follows a report this week of Chinese construction on four uninhabited features in the disputed Spratly islands, news that Beijing has dismissed as “unfounded”.

Any encroachment or reclamation on features within the Philippines’ 200-mile exclusive economic zone “is a threat to the security of Pagasa island, which is part of Philippine sovereign territory,” the ministry said in a statement, using the Filipino name for Thitu island.

“We strongly urge China to uphold the prevailing rules-based international order and refrain from acts that will exacerbate tensions,” it added.

The Chinese embassy in Manila reiterated that China strictly abides by a consensus reached among claimants that included not developing uninhabited reefs and islands.

Asked to respond to the defense ministry’s statement, it said both countries would “properly handle maritime issues through friendly consultations.”

China claims most of the South China Sea, through which billions of dollars’ worth of goods pass each year. Brunei, Malaysia, the Philippines, Taiwan and Vietnam also have overlapping claims to various islands and features.

Thitu is the most strategically important of nine features the Philippines occupies in the Spratlys, located close to Subi Reef, one of seven artificial islands that China has built on submerged reefs, some with surface-to-air missiles, aircraft hangars and runways.

Vietnam, Indonesia wrap up talks on exclusive economic zones

Vietnam and Indonesia have concluded negotiations on the boundaries of both countries’ exclusive economic zones (EEZ), a move praised by regional analysts but one likely to irk China.

“After 12 years of intensive negotiations, Indonesia and Vietnam have finally concluded negotiations on the EEZ boundaries of the two countries based on the 1982 UNCLOS,” said Indonesian President Joko Widodo after meeting with his visiting counterpart Nguyen Xuan Phuc on Thursday at the Bogor Presidential Palace in West Java.

UNCLOS is the abbreviation of the 1982 United Nations Convention on the Law of the Sea – a document to which Vietnam, Indonesia and China are signatories.

Phuc is in Indonesia on his first state visit to the Southeast Asian neighbor since taking office in 2021.

Vietnam and Indonesia have been embroiled in disputes over overlapping EEZ claims in the waters surrounding the Natuna Islands in the South China Sea for years.

Law enforcement agencies of the two countries have clashed over Vietnamese fishermen’s activities in the area, with Indonesia detaining and destroying dozens of Vietnamese boats accused of unlawful encroachment and illegal fishing.

The two countries agreed on a continental shelf, or seabed, limit in 2003 and since 2010, they have engaged in more than a dozen rounds of talks on EEZ delimitation. An EEZ gives a state exclusive access to the natural resources in the waters and in the seabed.

In 2021, Indonesian Minister of Foreign Affairs Retno Marsudi said in a statement that negotiating with Vietnam on EEZ borders in accordance with international law including UNCLOS 1982 was one of her ministry’s priorities.

“Indonesia will continue to reject claims that are not based on international law,” Marsudi said.

Important milestone

The news of the conclusion of negotiations was welcomed by regional observers and analysts who said it was an important achievement for both countries.

“It is a significant milestone,” said Shahriman Lockman, a director at Malaysia’s Institute of Strategic and International Studies (ISIS).

“It’s a big step towards having clearer demarcations of maritime boundaries between Southeast Asian countries,” Lockman said.

Nguyen The Phuong, a Vietnamese security analyst and a Lecturer at Ho Chi Minh City University of Economics and Finance, said the move “proves that ASEAN countries can settle maritime disputes amongst them.”

“It will help cool things off, especially issues relating to illegal, unreported and unregulated (IUU) fishing, which is a hot issue between Vietnam and Indonesia,” Phuong told RFA.

“It will also encourage the current negotiations between Vietnam and the Philippines, as well as Malaysia,” he added.

The South China Sea is being claimed by six parties – Brunei, China, Malaysia, the Philippines, Vietnam and Taiwan. Each party has its own set of maritime boundaries but Beijing’s claims are the most expansive.

Details of the newly reached Indonesia-Vietnam agreement have not been announced but their EEZ claims lie within China’s so-called “nine-dash line” that Beijing uses to demarcate its “historical rights” over almost 90% of the South China Sea.

“China would insist that they have jurisdiction over these areas,” according to Lockman from Malaysia’s ISIS.

A UN tribunal in 2016 invalidated China’s “nine-dash line” but Beijing has so far rejected the ruling, calling it “null and void.”

China is likely to voice protest against the new Indonesia-Vietnam agreement, warned Lockman and other analysts.
